卵巢癌(OC)是一种妇科肿瘤疾病,通常在晚期得到诊断,并且预后不良。已经确立了癌细胞的葡萄糖代谢速率显著高于正常细胞,而磷酸潘古糖途径(PPP)是葡萄糖代谢的一个重要支路。葡萄糖-6-磷酸脱氢酶(G6PD)是PPP中的关键限速酶,它在癌(如OC)的发生和发展中起着重要作用,并被认为是一种有前景的抗癌靶标。本综述根据G6PD的结构和生物功能,总结了G6PD在OC的进展、转移和耐药性中的作用的最新研究,并提出了可能的分子机制,这可能为针对OC患者的治疗提供了系统的理解。越来越多的证据表明,G6PD是癌症的一个有前景的靶标。开发G6PD抑制剂用于癌症治疗具有广泛的应用前景。
Ovarian cancer (OC) is agynecological tumor disease, which is usually diagnosed at an advanced stageand has a poor prognosis. It has been established that the glucose metabolismrate of cancer cells is significantly higher than that of normal cells, and thepentose phosphate pathway (PPP) is an important branch pathway for glucosemetabolism. Glucose-6-phosphate dehydrogenase (G6PD) is the key rate-limitingenzyme in the PPP, which plays an important role in the initiation anddevelopment of cancer (such as OC), and has been considered as a promisinganti-cancer target.In this review, based onthe structure and biological function of G6PD, recent research on the roles ofG6PD in the progression, metastasis, and chemoresistance of OC are summarizedand accompanied by proposed molecular mechanisms, which may provide a systematicunderstanding of targeting G6PD for the treatment of patients with OC.Accumulatingevidence demonstrates that G6PD is a promising target of cancer. Thedevelopment of G6PD inhibitors for cancer treatment merits broad applicationprospects.
